适用于Nimble Cloning系统的pCambia载体改造

论文摘要

为了适用于Nimble Cloning这种新型的DNA分子克隆技术,本研究以pCambia1304载体作为改造对象,采用突变碱基的方式去除载体中的5个Sfi I酶切位点,并且对改造后的载体在农杆菌中的稳定性以及在植物中的表达进行分析,结果表明5个Sfi I位点突变对载体的稳定性和表达没有影响。本研究为Nimble Cloning系统提供了配套载体,也为其他pCambia系列载体改造成Nimble Cloning系统载体提供了模板和参考。
